Default hygrometer at 75%

CC 48v 2000, Regen 2
Some of the cells on the batteries read full charge on the hygrometer, while other cells on the same battery read 75%. What does this mean tell me? Are new batteries in my future?
Fluid is full on all cells. Haven't load tested them ever.
This is my first cart.

A variation of 25% is beyond serviceable limits. My bet is a proper load test machine would show significantly diminished run time. Batteries are in your future.

I agree that your batteries are failing if you charged them before testing.
How old is your battery pack?
Also a hygrometer is for measuring barometric pressure and a hydrometer is for specific gravity.

The cart was refurbished and the previous owner told me the batteries where new. "Workoholics" by Interstate. The date stickers where not marked so I have no idea how old they are.
It seems odd to me that 3 cells test 100% and only one cell on each battery tests 75% and the weak cell seems to be on the end.

Weak cells can appear anywhere, I would try recharging the cart a couple of times to see if they come up.
If you are losing run time, it might be a good time to have the batteries load tested.
If you can't load test them, charge the cart, let it sit overnight, measure the individual battery voltages and post the results.
Then take the cart for a drive until the batteries start getting weaker, let the cart sit for a couple of hours and measure the individual battery voltages and post the results.
The members here will then have a good indication if your batteries are a problem or not and be able to offer some advice.
