ya think ? ...lol ... i would !!
also it is good to match batteries to with in .5v so they are equal in the charging cycle
if you put 3 good and 3 bad in pack the good will be brought down to the bads

No you do not need to disconnect any cables to test each battery. 6 volt batteries should never be run lower than 6.05 volts of a fully charged battery, and a 36 volt pack never below 36.3 that's 50% of the volts, when a battery or the pack is at 50% it needs to be charged
