May I ask if you opened up the intake or exhaust after boring out the cylinder?

I'm going on a basis of a lifetime of experience with numerous motors, so I may be making a leap in the wrong direction, but I think you will need to balance the intake and exhaust to match the larger bore of your motor. What you are describing sounds like what used to be called "Vapor Lock". Basically you are creating a vacuum that can't suck in enough air through the intake to fill the void. Also on exhaust you would find the piston working too hard to expel the gases due to the restriction. That's my informed guess as to why you can get the piston to move without the spark plug in the head.

Here's something to help you see the process. http://science.howstuffworks.com/tra...wo-stroke2.htm

If you watch the animation you might get other ideas about what's wrong.
