yes if its a 48 volt cart and they are good 12 volt batteries then yes they will drive the cart...................................

Also, will the 12v car batteries damage the cart motor if used?

they will not damage motor but keep in mind if your gonna throw car batteries in it i hope its just for testing purposes......because they arn,t gonna get you anything as far as drivetime ..............they arn,t made to handle the voltage or amps demands but for testing and very very short drive they,ll work.........please tell us your not putting car batteries in to use for driving the cart?? TESTING PURPOSES ONLY................................

Low end 12v batteries would be fine for testing . . but for price of 4 batteries to test motor, why not just take motor out and to a shop for them to bench test it?
